Cagrilintide
Also known as AM833, CagriSema (combined with semaglutide)
Cagrilintide is a long-acting amylin analog that targets appetite and satiety via a different pathway than GLP-1 agonists. Its key application is CagriSema — a fixed-dose combination with semaglutide 2.4mg — which showed ~22.7% weight loss in Phase 2 by targeting both amylin and GLP-1 pathways simultaneously. The complementary mechanisms produce additive weight loss beyond either drug alone.
How it works
Activates amylin receptors (AMY1–AMY3) in the hypothalamus and brainstem to directly reduce appetite and caloric intake via the circumventricular organs — a distinct pathway from GLP-1 incretin signaling. Complementarity with semaglutide creates additive appetite suppression.

Dosing
0.3–4.5 mg · Once weekly
Titration: 0.3 → 0.6 → 1.2 → 2.4 → 4.5mg (every 4 weeks). CagriSema Phase 3 dose: 2.4mg cagrilintide + 2.4mg semaglutide once weekly.
