VIP (Vasoactive Intestinal Peptide)

How it works

Binds VPAC1 and VPAC2 receptors (coupled to adenylyl cyclase), activating cAMP-PKA signaling that suppresses NF-κB and reduces pro-inflammatory cytokine production (TNF-α, IL-6, IL-12). Promotes regulatory T-cells (Tregs), modulates mast cell degranulation, and exerts direct neuroprotective effects in CNS.

Dosing

50–100 mcg · 4x daily (nasal) or once daily (SubQ)

Nasal spray: 50mcg per nostril 4x daily (200mcg/day total). SubQ: 50–100mcg once daily. Nasal route preferred per Shoemaker protocol. Do NOT use in CIRS until MARCoNS cleared and all prior protocol steps complete.

Reconstitution

For nasal spray: reconstitute in preserved saline for nasal use. 10mg vial typically diluted to 50mcg per pump. Requires nasal atomizer.

Who it’s for

Patients with CIRS/biotoxin illness (must be the final step after completing Shoemaker Protocol). Those with MCAS, chronic inflammatory conditions, or severe neuroinflammation. Advanced use — not recommended without functional medicine supervision.
